芳香化酶抑制剂对胶原性关节炎大鼠血清中IL-17和RANKL／RANK／OPG的影响

摘　　要：目的探讨第三代芳香化酶抑制剂阿那曲唑对胶原诱导的关节炎（ClA）大鼠血清中自介素17（IL—17）、核因子-KB受体活化因子（RANK）及其配体（RANKL）和骨保护素（OPG）的作用，了解其加重骨关节病变作用及其机制。方法建立C1A大鼠模型，分为阿那曲唑组、卵巢切除组和假手术组，用排水法大鼠关节炎足体积变化，放射线分析大鼠关节病变情况，并用ELISA法分析C1A大鼠血清中IL-17、RANKL及RANK和OPG的表达情况。结果与卵巢切除组比较，阿那曲唑组大鼠关节病变加重（P〈0．05），血清中IL－17、RANKL表达增加，OPG的表达显著降低，RANKL／OPG比值明显增加，差异均具有统计学意义（P〈0．05）。结论在CIA大鼠模型中，阿那曲唑可增加大鼠衄清中IL－17、RANKL的表达，而抑制OPG的表达，导致大鼠关节病变加重。Objective To explore the effect of aromatase inhibitors - anastrozole on IL - 17 and RANKIJRANK/OPG in the serum of collagen induced arthritis （ CIA ） rats and to illustrate its aggravation on bone. Methods CIA rats were divided randomly into 3 groups：anastroznle group（A）, ovarectomy group（O） anti sham operation group（S）. The edema rate of ankle joints were examined. The radiological injuries of ankle were observed by molybdenum target radiography. The production of IL - 17 and RANKL/RANK/OPG in the serum were determined by enzyme linked immunosorbent assay（ELISA）. Results Compared to ovarectomy group, ad- ministration of anastrozole significantly increased the severity of al^hritis （ P 〈 O. 05 ）, the levels of IL - 17 and RANKL in anastrozole group were significantly higher, whereas, OPG expression was lower. Moreover, the ratio of RANKL/OPG obviously increased （P 〈 0.05 ）. Conclusion Anastrozole potently promots the progression of CIA model by the increasing of IL - 17 ,RANKL and decreasing of OPG.
