多花水仙花朵全长cDNA文库的构建

摘　　要：采用SMART技术,以多花水仙‘黄花水仙2号’盛开期花朵为材料,构建其全长cDNA文库,并对文库质量进行鉴定.结果表明,所构建初始文库的滴度为6.30×105cfu·mL-1,扩增后文库的滴度为1.58×108cfu·mL-1,重组率为84%,插入片段大小集中在500-2000 bp之间,平均长度约970 bp,符合cDNA文库的质量标准.可见,所构建的文库比较完整,能够反映黄花水仙花朵盛开期基因的表达情况.The flowers of Narcissus tazetta L. cultivar Huanghua at full-bloom stage were used as material to construct a full-length cDNA library using switching mechanism at 5＇end of RNA transcript （SMART） strategy and the quality of the library was detected. The results show that the titer of the primary library was 6.30 × 10^5 cfu·mL^-1, the recombinant rate was 84%. The insert fragments concentrated in 500 - 2000 bp with an average size was about 970 bp. These results indicate that the cDNA library constructed in this report accords with cDNA library quality standards and is relatively complete. It can reflect the genes expression in full- blossom of N. tazetta cultivar Huanghua.
