摘  要:目的研究艾迪注射液对人肺癌H460细胞的放射增敏作用及机制。方法 MTT法测定药物对细胞的抑制作用;克隆形成法观察药物对细胞的放射增敏作用;流式细胞术分析药物对细胞周期分布的影响;Western blot法检测裂殖酵母cdc2基因编码蛋白(cdc2蛋白)在H460细胞中表达的变化。结果不同浓度艾迪注射液(3-192mg/ml)对H460细胞作用48h后均有生长抑制作用。艾迪注射液对H460细胞有明显的放射增敏作用,作用24h和48h后平均致死剂量增敏比(SERD0)分别为1.01和1.17,准阈剂量增敏比(SERDq)分别为1.07和1.37。流式细胞仪检测药物作用后G2期细胞增加。Western blot示单纯照射组细胞cdc2蛋白较对照组无明显减少,但药物与照射联合组cdc2蛋白表达量较单纯照射组下降(P<0.05)。结论艾迪注射液可以增加人肺癌H460细胞的放射敏感性,可能与对细胞G2期阻滞及减少cdc2蛋白表达有关。Objective To investigate the radiosensitivity enhancement effect and underlying mechanism of Aidi injection on lung cancer cell H460. Methods The H460 cells were treated with Aidi injection. The proliferation of cells was measured by MTT assay and cell survival was determined by clonogenic assay. Cell cycle distribution after Aidi injection treatment was analyzed by flow cytometry. Western blot was used to determine cdc2 protein expression. Results The inhibition rates of H460 cells were different with different concentrations of Aidi injection 3-192 mg/ml for 48 h. The cell survival experiment showed the sensitivity enhancement ratioes of 1.07 and 1.37 for 24 h and 48 h of Aidi injection. DNA flow cytometric analysis indicated that Aidi injection (12 mg/ml) induced efficiently G2 phase arrest following 48 h of exposure. Meanwhile the ode2 level was decreased in Aidi injection and radiation-treated H460 cells compared with control cells(P〈0. 05). Conclusion Aidi injection can enhance the radiosensitivity of H460 cells, which may be related to G2 phase arrest and downregulation of cdc2 expression.
