摘  要:目的通过检测胃肠道肿瘤患者血浆中活性氧类物质、抗氧化酶防御系统以及脂质过氧化物丙二醛(MDA)和DNA氧化损伤生物学标志物8-羟基脱氧鸟嘌呤(8-OHdG)的水平,分析胃肠道肿瘤患者机体氧化应激状态。方法采用紫外分光光度仪检测胃肠道肿瘤患者(胃癌41例,肠癌62例)以及40例正常人血清中谷胱甘肽过氧化物酶(GSH-PX)、总抗氧化能力(T-AOC)、超氧化物歧化酶(SOD)、MDA、超氧阴离子(O2-)、过氧化氢酶(CAT)水平;采用酶联免疫吸附法(ELISA)测定8-OHdG水平。结果与对照组比较,胃癌、肠癌组血清中O2-活力、MDA含量明显升高(P<0.05);T-AOC、SOD、GSH-PX、CAT明显降低(P<0.05);胃癌、肠癌组血清中8-OHdG明显升高(P<0.05);胃、肠癌组间无显著性差异。结论胃肠道肿瘤患者体内活性氧成分增高;抗氧化酶降低;氧化损伤产物增加,过高的氧化应激水平参与胃肠道肿瘤的发生。Objective To analyze the level of oxidative stress in gastrointestinal cancer by detecting reactive oxygen species (ROS), antioxidant enzymes, and oxidative damage products such as lipid peroxidation product and the biomarker of oxidative DNA damage (8-OHdG). Methods The plasma levels of glutathione peroxides (GSH- PX), total antioxidative capability (T-AOC), superoxide dismutase ( SOD), malondialdehyehyde ( MDA), superoxide anion (02- ) and hydrogen peroxidease (CAT) were determined by colorimetry, and the plasma levels of 8-OHdG were determined by enzyme-linked immunosorbent assay (ELISA) in 41 gastric cancer and 62 intestinal cancer patients as well as in 40 healthy subjects. Results Compared with the healthy controls, patients with gastrointestinal cancer had significantly increased plasma levels of 02- and MDA (P(0.05) and decreased plasma GSH-PX, T-AOC, SOD and CAT levels (P〈0. 05). The plasma level of 8-OHdG was significantly higher in patients with gastrointestinal cancer than in the healthy controls (P〈 0. 05). ConcEusion Patients with gastrointestinal cancer have increased levels of ROS, decreased levels of antioxidant enzymes and accumulated oxidative damage products. High level of oxidative stress is involved in gastrointestinal cancer.
