摘 要:目的观察普罗布考对血管性痴呆(vascular dementia,VD)模型大鼠学习记忆改善情况及海马区SOD和MDA的影响。方法将SD大鼠随机分为模型组、假手术组、普罗布考组。双侧颈总动脉永久性结扎法制作VD大鼠模型,给药8w后以Morris水迷宫实验观察大鼠学习记忆情况。用HE染色观察海马神经元损伤情况,并测定海马区SOD活力及MDA含量。结果与假手术组相比,模型组大鼠学习记忆能力明显减弱(P<0.01);海马区SOD活性降低,MDA含量明显增加(P<0.01)。与模型组相比,普罗布考组大鼠学习记忆能力明显改善(P<0.01),海马区SOD活性明显增加,MDA含量明显降低(P<0.01);海马CA1区神经元损伤明显改善。结论普罗布考可改善VD大鼠认知水平和病理变化,其保护作用可能与提高SOD活力、降低MDA含量从而减少脂质过氧化有关。Objective To study the effects of probucol on cognitive function and the change of Superoxide Dismutase (SOD) and Malondialdehyde (MDA) of hippocampns in vascular dementia (VD) model rats. Methods Sprague-Dawley (SD) rats were randomly divided into model group, sham-operated group and probucol group, VD model was set up and pro- bucol was administered for 8 weeks. Morris water maze test was established to detect the ability of learning and memory of rats. Hippocampal neuron injury was observed by HE staining, and the level of SOD and MDA were measured by zanthine oxidase method and Thibabituric Acid (TBA) method respectively. Results Compared with the sham-operated group, learning and memory ability of rats in the model group decreased significantly( P 〈 0.01 ), SOD activity in the hippocampus decreased significantly and the content of MDA significantly increased( P 〈 0.01 ). Compared with the model group, learning and memory ability of rats in probucol treatment group improved significantly(P 〈 0.01 ) ,the SOD activity in hippocampus obviously increased and MDA content significantly decreased ( P 〈 0.01 ). Conclusion Probucol can improve cognitive lev- el and pathological changes in VD rats. The mechanisms may be related to reduce lipid peroxidation by increasing the activi- ty of SOD and decreasing content of MDA.
