摘  要:目的观察盐酸多奈哌齐治疗的血管性痴呆(VD)大鼠海马CA1区神经细胞凋亡及突触素(synaptophysin,Syp)的变化,探讨其在VD治疗中的作用机制。方法采用永久性结扎双侧颈总动脉法制作大鼠VD模型,将60只SD大鼠随机分为假手术组、手术对照组、药物治疗组各20只,用免疫组织化学染色方法检测大鼠海马CA1区Syp表达,用TUNEL染色检测神经细胞凋亡。结果治疗组大鼠海马CA1区Syp表达明显高于手术对照组;海马区神经细胞凋亡明显少于手术对照组(P均<0.01),与假手术组比较均无统计学差异(P>0.05)。结论多奈哌齐可使Syp表达增加,使突触重塑,这可能改善实验动物的神经行为能力、达到改善VD大鼠认知能力的作用。Objective To investigate the mechanism of action of donepezil in vascular dementia by study the change of Synaptophysin(Syp) expression and neuron apoptosis in hippocampal CA1 of vascular dementia rats. Methods 60 SD rats were randomly devided into three groups:sham operation group, VD model group and Donepezil treated group, with 20 rats in each. VD model were established by occlusion of the bilateral common carotid arteries. The expression of Syp in hipp- ocampal CAI were measured by immunohistochemical staining and neuron apoptosis was observed with terminal deoxynucle- otidyl transferase mediated dUTP-hiotin nick end labeling (TUNEL) staining. Results In comparison with VD model group, the expression of donepezil treated group of Syp in hippocampal CA~ increased significantly (P 〈 0. O1 ) ;neuron apop- tosis in hippocampal decreased significantly( P 〈0.01 ). Donepezil treated group with no statistical difference in both of ex- pression of NMDAR-2B and neurons apoptosis compared with the sham operation group. Conclusion Donepezil is able to promote remodeling of new synapses, safeguard neurons and improve impaired learning and memory ability in upregulating expression of Syp in hippocampal CAl of VD rats.
