摘  要:目的研究辽东楤木叶总皂苷(ETS)对HT-29细胞生长的抑制作用及作用机制。方法采用MTT法检测ETS对HT-29细胞增殖的抑制作用;流式细胞仪检测ETS对HT-29细胞周期和凋亡的影响;Western blot法检测ETS对HT-29细胞PLK1蛋白表达的影响。结果 ETS对结肠癌HT-29细胞增殖有明显抑制作用,IC50值为39.62 mg·L-1。在10~40 mg·L-1剂量作用下,可剂量依赖性诱导HT-29细胞凋亡。ETS主要将细胞周期阻滞在G0/G1期,从而可剂量依赖性抑制肿瘤细胞增殖。ETS可明显抑制HT-29细胞PLK1蛋白的表达。结论 ETS体外可明显抑制HT-29细胞的生长,其抗肿瘤作用与细胞周期阻滞、细胞凋亡及抑制PLK1蛋白表达有关。Aim To investigate the growth inhibitory effects of total saponins from leaves of Aralia elatl Seem (ETS) on human colon cancer cells and explore the related mechanism. Methods The inhibitory rate of ETS on HT-29 cell proliferation was determined by MTF method. Annexin V-FITC and PI staining were used to examine cell apoptosis and cycle. The expression of PLK 1 protein was analyzed by Western blot method. Results Cell proliferation was inhibited in HT-29 cells treated with ETS, and the value of IC50 was 37.39 mg· L^-1 after 48 h. ETS could induce HT-29 cells apoptosis and arrest cell at G0/G1 phase, while decreasing the expression of PLK1 protein. Conclusions ETS can inhibit the proliferation of human colon cancer cell, and the mechanism may be related with the induction of cell apoptosis, the change of cell cycle (G0/G1 phase arrest) and the down-regulation of the expression of PLK1 protein.
