核基质结合区结合蛋白1在结直肠癌细胞中的表达及其临床意义

摘　　要：目的探讨核基质结合区结合蛋白1(SATB1)在结直肠癌细胞中的表达及其临床意义。方法以大肠癌细胞株SW480、SW620为研究对象,运用免疫细胞化学分析SATB1在细胞内的表达情况。将142例标本分为癌旁组织、腺瘤组织、大肠癌组织、淋巴结转移癌,运用免疫组织化学方法分析SATB1在这些组织中的表达情况。结果 SATB1在人大肠癌细胞系SW480与SW620细胞中均有较强的表达,并且定位在SW480与SW620细胞的细胞核。SATB1在正常结直肠粘膜、腺瘤、癌及淋巴结转移癌组织中的表达明显不同,差异有统计学意义(χ2=79.653,P<0.05)。SATB1表达与结直肠癌转移高度相关(Z=-2.115,P<0.05),即SATB1表达弱者转移的发生率低,表达强者其转移发生率高。结论SATB1在结直肠癌细胞中高表达,与结直肠癌发生演进及转移存在联系。Objective To study the association between special AT-rich sequence-binding protein 1 （SATB1） and colorectal cancer（CRC）. Methods The expressions of SATB1 in SW480 and SW620 cells were detected by immunocytochemistry technique,and the protein expression level of SATB1 was also measured by immunohistochemical technique in samples from patients of colorectal carcinoma at different stages.Normal mucosa of colon and colonic adenoma were used as controls. Results The SATB1 protein was mainly located in cell nucleus of SW480 and SW620. Protein expression levels of SATB1 in colorectal carcinomas,normal mucosa,adenoma and metastatic lymph nodes were significantly different （x2=79.653,P〈0.05）. There was correlation between the expression level of SATB1 and progression of colorectal carcinoma including metastasis （Z=-2.115,P〈0.05）. Conclusion Our results indicated that SATB1 may play an important role in the progression of human colorectal cancer.
