摘 要:以榨菜B2-1014茎尖和下胚轴为外植体,采用不同浓度的秋水仙素处理,培养不同时间,通过根尖染色体计数法及气孔观察法对诱变植株进行倍性鉴定,获得四倍体植株。结果表明,不同处理条件可以产生不同的诱导效果;同样条件下以茎尖为外植体诱导效果较好;最佳处理条件组合为秋水仙素浓度0.4%、培养时间48 h,四倍体加倍率达最高,为30%;与原二倍体榨菜相比,四倍体榨菜植株具有叶片变大、茎变粗、球茎变大等特点。In order to obtain tetraploid tuber mustard, stem tip and hypocotyledonary axis of cultivar 'B2-1014' were used as explants and treated with colchicines of different concentrations and treatment times. Ploidy identification of mutagenic plants adopted the means of root tip chromosome counting and stoma observation. The results indicated that different processing conditions had diverse induction effect. Using stem tip as explants was better than hypocotyledonary axis as explants. The optimum processing condition was 0.4% to concentrations of colchicines and 48 h to treatment times. Under this treatment, tetraploid plants inducement rate was 30%, which was the highest in all the treatments. Compared with the original diploid plants of tuber mustard, tetraploid plants had charac- teristics of larger leaves, thicker stems and larger bulbs.
