摘 要:目的 研究锌对人胚大脑皮质神经细胞增殖周期 DNA和蛋白质含量的影响。方法 在建立原代培养的人胚大脑皮质神经细胞模型的基础上 ,采用免疫组织化学染色和流式细胞术进行研究。结果 (1 )锌促进神经细胞的存活作用具有剂量效应关系。 0 .2~ 4.0 mg/L锌实验组神经细胞存活数显著高于对照组 (P<0 .0 1 ) ,且 2 .0 mg/L锌实验组为最佳剂量组。6.0 mg/L锌组神经细胞存活数显著低于对照组 (P<0 .0 1 )。(2 ) 7、1 4和 2 1 d锌实验组神经细胞 DNA和蛋白质含量均显著高于对照组 (P<0 .0 1 )。 (3)锌实验组的增殖指数、S和 G2 +M期细胞百分率高于对照组 ,G0 /G1期细胞百分率则低于对照组 (P<0 .0 1 )。结论 一定剂量范围内的锌能促进神经细胞增殖。Objective: To investigate the effect of zinc on the content of DNA and protein of cerebral cortex neurons of human fetus in vitro. Methods: FACS and stained immunohistochemical methods were used. Results: (1) There was dose dependent effect of zinc on the neuron survival. 0.2~4.0 mg/L zinc could significantly promote neuron survival. 2.0 mg/L zinc was the best concentration of the incubation fluid. In the presence of 6.0 mg/L zinc, the growth and development of neurons were inhibited. (2) The total DNA and protein contents in zinc group were enhanced significantly as compared with control. (3) The proliferative index and the proportions of S and G 2+M phase cells in zinc group were markedly higher than control. But G 0/G 1 phase cells were significantly lower than control. Conclusion: Zinc in certain range of concentrations of the incubation fluid can promote the neurons proliferation and the synthesis of DNA and protein in vitro.
