摘  要:目的 探讨BRCA1在家族性乳腺癌中的作用及其与p5 3的相关性。方法 对家族性乳腺癌手术标本及非家族性乳腺癌手术标本共 84例进行BRCA1及 p5 3蛋白免疫组化 (S P法 )检测。 结果 BRCA1在家族性乳腺癌中及非家族性乳腺中的阳性率分别为 6 1.9%和 11.9% ,p5 3在家族性乳腺癌及非家族性乳腺癌中的阳性率分别为 40 .5 %和 2 6 .2 % ,而在家族性乳腺癌BRCA1阳性病例中有 5 7.7%的病例 p5 3阳性 ,在非家族性乳腺癌BRCA1阳性病例中仅 40 .0 %的病例 p5 3阳性。 结论 BRCA1突变在家族性乳腺癌的发生过程中具有重要作用 ,BRCA1突变与 p5 3突变具有显著相关性。p5 3突变可能为BRCA1突变的 1个步骤。Objective To explore the role of the breast cancer susceptibility gene BRCA 1 in familial breast cancer and the relation between BRCA 1 and p53.Methods The levels of BRCA 1 and p53 protein were detected by immunohistochemistry in 84 breast cancer paraffin-embeded blocks.Results The rate of BRCA 1 expression in familial breast cancer and non-familial breast cancer was 61.9% and 11.9% respectively.The rate of p53 expression in familial and non-familial breast cancer was 40.5% and 26.2%.In familial breast cancer cases with BRCA 1 expression,the rate of p53 expression was 57.7%,while non-familial breast cancer with BRCA 1 positive reaction,it was only 40.0%.Conclusion The mutation of BRCA 1 plays a major role in familial breast cancer.And the mutation of BRCA 1 have relation to the mutation of p53.The mutation of p53 is probably a necessary step in the mutation of BRCA 1.
