摘 要:目的研究姜黄素干预小鼠载脂蛋白B100(ApoB100)和固醇调节元件结合蛋白1c(SREBP-1c)在肝脏表达与非酒精性脂肪性肝病(NAFLD)的相关性,从基因水平探讨姜黄素治疗NAFLD的机制,为姜黄素的临床应用提供基础理论依据。方法应用实时PCR方法对比正常组、NAFLD模型组、姜黄素预防组、姜黄素治疗组中肝脏ApoB100和SREBP-1c的mRNA表达进行分析。结果 ApoB100 mRNA在模型组表达增高,而在预防组和治疗组表达均降低(P<0.05);SREBP-1c mRNA在模型组表达增高,而在预防组和治疗组与正常组相比表达均增高(P<0.05)。结论应用姜黄素对于NAFLD的预防和治疗作用可能不是通过调节SREBP-1c mRNA的表达而实现的。Objective To investigate the relationship between apolipoprotein B100 (ApoBl00) and sterol-regulatory element binding protein-1 c (SREBP-1c) and nonalcoholic fatty liver disease (NAFLD) in liver of mice after interference by curcumin.Methods Expressions of ApoB100 and SREBP-1c mRNA in liver of normal,NAFLD model,and curcumin prevention Treatment groups were tested by RT-PCR.Results The expression of ApoB100 mRNA was increased in NAFLD model group,and decreased in prevention group and treatment group (P < 0.05).The expression of SREBP-1 c mRNA was increased in NAFLD model group,and also increased in prevention group and treatment group compared with normal group (P < 0.05).Conclusions The expression of ApoB100 mRNA is increased in NAFLD model group,and curcumin controlls the lipid metabolism by suppressing the expression of ApoB100 mRNA.The expression of SREBP-1c mRNA is increased in NAFLD model group,prevention group and treatment groups,and curcumin maybe treat NAFLD by other way instead of affect the expression of ApoB100 mRNA.
