摘　　要：目的 :研究白介素 - 2 (interleukin- 2 ,IL - 2 )基因转染的人卵巢癌细胞对人外周血淋巴细胞 (peripheral bloodiymphocytes,PBL)杀瘤活性的影响。方法 :采用脂质体携带 IL- 2基因转染人卵巢癌细胞系 3AO细胞 ,与正常人 PBL 混合培养 ,利用 MTT法测定混合培养后 PBL对已转染 IL- 2基因的 3AO细胞抑制率。结果 :基因转染后 3AO细胞生长良好 ,光镜形态下未观察到凋亡改变 ;转染后的 3AO细胞转录并产生 IL - 2 m RNA;PBL对转基因 3AO细胞的抑制率明显高于无基因转染的 3AO细胞的抑制率 (P<0 .0 1)。结论 :IL- 2基因修饰的卵巢癌细胞能够改变淋巴细胞的功能而发挥其抗肿瘤作用。Objective:To investigate the effects of interleukin 2(IL 2) gene transduced human ovarian cancer cells on the cytotoxity of human peripheral blood lymphocytes(PBLs).Methods:IL 2 gene was transfected into human ovarian cells 3AO,then it was cocultured with PBLs for 48 hours.Cytotxicity of PBLs on 3AO cells was detected by MTT assay.Results:Morphology of IL 2 gene transduced 3AO cells remained unchanged.IL 2 gene could be transfected into the 3AO cells,IL 2 gene transduced 3AO cells could expressed IL 2mRNA and enhanced significantly their response to cytotoxicity of PBLs( P <0.01).Conclusion:IL 2 gene transduced ovarian cancer cells can enhance their response to cytotoxicity of PBLs,it may be a useful gene therapy for the ovarian cancer.
