溃疡性结肠炎患者血浆及肠黏膜组织中IL-23、IL-17的表达及相互关系

摘　　要：目的:检测白介素-23(interleukin-23,IL-23)和IL-17在溃疡性结肠炎(ulcerative colitis,UC)患者血浆及肠黏膜组织中的表达,并探讨二者间关系.方法:留取40例UC患者血浆及肠黏膜组织标本,进行组织学分级,同时收集12例正常对照者血浆和肠黏膜组织标本.ELISA法检测血浆中IL-23、IL-17水平,免疫组织化学SP法检测肠黏膜组织中IL-23、IL-17的表达,并分析两者关系.结果:40例UC患者组织学分级1级7例,2级21例,3级12例.UC患者血浆及肠黏膜组织中IL-23、IL-17表达均高于正常对照组(分别为:0.17±0.58 vs 4.38±1.27,P<0.05;0.42±1.00vs 4.88±0.91,P<0.05),且二者的表达随UC组织病理分级增加而增高(分别为:3.00±0.53vs4.12±1.15 vs 5.58±0.51;3.71±0.76 vs 4.81±0.60 vs 5.67±0.65),IL-23与IL-17呈显著正相关(P<0.01).结论:IL-23、IL-17在UC的病程中可能发挥重要作用.AIM： To determine the levels of interleukin 23 （IL-23） and IL-17 in the plasma and colonic mucosa in patients with ulcerative colitis （UC） and to analyze their correlations. METHODS： ELISA and immunohistochemistry were used to determine the levels of IL-23 and IL-17 in the plasma and colonic mucosa in 40 UC patients and 12 healthy controls, respectively. RESULTS： Both plasma and colonic mucosal levels of IL-23 and IL-17 were significantly increased in UC patients compared to healthy controls （all P 〈 0.05）. IL-23 and IL-17 immunoreactivity was significantly associated with pathologic grade of UC. The expression of IL-17 had a positive correlation with that of IL-23. CONCLUSION： IL-23 and IL-17 may play an important role in the progression of UC.
