摘  要:目的:研究血红素氧合酶-1(heme oxygenase-1,HO-1)在食管鳞癌侵袭转移中的意义。方法:采用免疫组化方法检测HO-1在114例食管鳞癌组织中的表达水平;细胞划痕实验验证高低转移食管鳞癌细胞体外侵袭转移能力的差异;Western blot及荧光定量PCR(qRT-PCR)检测HO-1在两株食管鳞癌高低转移细胞系中的表达差异。结果:免疫组化结果提示HO-1在食管癌组织细胞质和细胞膜呈高表达,其阳性表达率与肿瘤分化程度(P=0.039)、浸润程度T分期(P=0.020)、淋巴结转移(P=0.017)、淋巴管浸润(P=0.037)、远处转移(P=0.038)有显著相关性;细胞划痕实验验证了食管鳞癌高低转移细胞侵袭转移能力的差异,qRT-PCR及Western blot结果提示HO-1 mRNA及蛋白在两株高转移食管鳞癌细胞株表达水平均显著高于对应的低转移细胞株。结论:HO-1在食管癌组织及高转移性细胞系中呈高表达,提示HO-1和食管鳞癌的侵袭转移相关。Objective:To evaluate the significance of HO-1 expression in the invasion and metastasis of human esophageal squamous cell carcinoma (ESCC).Methods:Immunohistochemistry was used to evaluate the level of HO -1 expression in 114 ESCC samples and the corresponding adjacent tissues.Metastatic ability of the ESCC cells was tested by wound-healing assays.Western blot and quantitative real-time PCR (qRT-PCR) were performed to identify the differences of HO-1 expression between two highly metastatic and non-metastatic ESCC cell lines.Results:HO-1 expression was found to be positively stained in the cell cytoplasm and cell membrane of ESCC tissues.Statistical analyses showed that the expression of HO-1 in ESCC significantly correlated with the patient TNM stage (P =0.020),lymph node metastasis (P =0.017),lymphatic vessel invasion (P =0.037) and distant metastasis (P =0.038).Wound-healing assays showed that highly metastatic cells had a better metastatic ability than lowly metastatic cells.qRT-PCR and Western blot analyses revealed that the expressions of both mRNA and protein of HO-1were markedly higher in the highly metastatic cells compared with the non-metastatic cells.Conclusion:The expression of HO-1 is significantly increased in ESCC tissues and highly metastatic cell lines,indicating that HO-1 plays an important role in the metastasis of ESCC and might provide a potential prognostic and therapeutic marker for ESCC patients.
