摘　　要：目的　将表达IL 2重组BCG用于动物实验 ,观察其对肿瘤生长和机体免疫功能的影响。方法　采用基因工程技术和电转化技术 ,构建重组卡介苗即rBCG IL 2和rBCG ,并用瘤株重组BCG接种荷瘤小鼠 ,几周后观察肿瘤生长情况和检测巨噬细胞的杀伤活性。结果　rBCG IL 2能表达分泌IL 2 ,其表达量为 9 87ng ml± 4 56ng ml,rBCG IL 2与rBCG抑制肿瘤生长的作用明显优于对照组 ,并能提高小鼠巨噬细胞杀瘤率。结论　rBCG IL 2能有效抑制肿瘤生长 ,较早刺激机体的免疫功能 ,提高巨噬细胞的杀瘤活性。Objective To inquire into the effect of rBCG-IL-2 on tumor growth and immunity after it has been inoculateded in animal.Methods Gene engineering and electric transformation technologies were adopted to construction recombinant BCG,one of which was rBCG-IL-2 strain and the other was rBCG strain.Observe the growth of tumor and determine tumoricidal effect of macrophage after the tumor cells and recombined BCG have been inoculated in mouse in a few weeks.Results rBCG-IL-2 expressed and secreted IL-2 and the count of secreting was about 9.87ng/ml±4.56ng/ml.The anti-growth effects of tumor of rBCG-IL-2 and rBCG were obvious higher than the control.Conclusion rBCG-IL-2 can prevent tumor from growing effectually,active immunity of the host early and enhance cytotoxicity of macrophage obviously.
