摘  要:目的:研究米力农对兔蛛网膜下腔出血(SAH)后脑血管痉挛的作用及潜在机制。方法:建立兔SAH模型。采用CT血管成像(CTA)和经颅多普勒超声(TCD)测量实验动物基底动脉的管径和血流速度。结果:SAH动物的基底动脉管径变细、血流速度增快。米力农给药组SAH动物平均基底动脉管径增粗而血流速度下降,而L-NAME(NOS阻断剂)可阻断米力农的药理作用。结论:米力农在下腔出血后脑血管痉挛的发展中起到重要作用。可能在一定程度上是通过增加eNOS的蛋白表达起到缓解脑血管痉挛的作用。Objective: To evaluated the effect of milrinone in a rabbit model of cerebral vasospasm and eek to identify the underlying mechanisms of action. Methods: A rabbit SAH model was established by double injection of autoblood via cisterna magna. Cerebral blood flow and arterial diameter were measured with the Transcranial Doppler (TCD) and Computed Tomography Angiography (CTA). Results: SAH induction resulted in significantly reduced diameters and accelerated blood flow velocities in basilar arteries in SAH - in- duced animals (P 〈0. 05 vs. sham group), whereas animals also treated with L -NAME (a NOS inhibitor) showed attenuation of the milrinone effect. Conclusions: Milrinone plays an important role in the post -SAH cerebral vasospasm. Milrinone had vasodilatory effects mediated at least partly by means of increasing of expression of eNOS levels.
