摘  要:目的:探讨海洛因依赖对大鼠中脑腹侧被盖区(VTA)β-内啡肽(β-endrophin,β-EP)表达的影响。方法:成年雄性SD大鼠55只,分为海洛因依赖组(HDG)及生理盐水对照组(SCG)各25只,正常对照组(NCG)5只,建立海洛因依赖模型;分别于第10、17、24、31、38天取脑组织,用免疫组织化学SABC法显示VTA区β?EP阳性细胞,并用图像分析法测定平均光密度值。结果:显微镜观察显示,与盐水对照组和正常对照组比较,海洛因依赖组VTA区的β?EP阳性细胞免疫反应强度减弱,染色变浅;图像分析测定大鼠VTA区β?EP阳性细胞的平均光密度值,盐水对照组与正常对照组比较差异无统计学意义(P﹥0.05),海洛因依赖组与正常对照组和盐水对照组比较均显著降低(P﹤0.05),且随着依赖时间延长呈下降趋势。结论:在海洛因依赖期间,VTA区β?EP表达减少,提示海洛因抑制了内源性阿片肽的产生。Objective: To investigate the (β-EP) in ventral tegmental area (VTA) effect of heroin dependence on expression of β-endrophin of rat brain. Methods: Fifty-five male adult SD rats were randomly divided into a normal control group (group NC, n = 5 ), a saline control group (group SC, n = 25) and a heroin dependent group ( group HD, n = 25 ). Heroin-dependent rat models were set up by subcutaneous injection of heroin and the brain tissue were excised on the 10^th, 17^th, 24^th, 31^th, and 38^th days. Immunohistochemical SABC method was used to detect the β-EP positive cells in VTA, and average optical density was examined by using imagine analysis method. Results:The immune reaction intensity in β-EP positive cells of group HD was weaker, and the color was lighter than those of groups NC and SC ( P 〈 0.05 ). The average optical density of β-EP positive cells was not significantly different between groups SC and NC, but itg lower in group HD than in the 2 groups, and showed a downtrend along with time ( P 〈 0.05 ). Conclusions: The expression of β-EP in VTA decreases during heroin dependence,which suggests that heroin inhibits the production of endogenous opioid.
