HPLC法同时测定大败毒胶囊中绿原酸、咖啡酸和阿魏酸的含量

摘　　要：目的:建立HPLC法同时检测大败毒胶囊中绿原酸、咖啡酸和阿魏酸的测定方法。方法:采用Agilent C_(18)(250mm×4.6mm,5μm)色谱柱;乙腈-0.4%磷酸溶液(15:85)为流动相,流速:1.0ml·min^(-1);检测波长:324nm。柱温:30℃,进样量:10μl。结果:线性范围:绿原酸5.42~32.43μg·ml^(-1)(r=0.9998),咖啡酸2.63~26,32μg·ml^(-1)(r=0,9994),阿魏酸1.44~14.44μg·ml^(-1)(r=0.9999);平均回收率:绿原酸99.98%,RSD=0.2%(n=6),咖啡酸99.31%,RSD=0.4%(n=6),阿魏酸99.48%,RSD=0.8%(n=6)。结论:该方法操作简便、准确,重复性好,可用于测定大败毒胶囊申绿原酸、咖啡酸和阿魏酸的含量。Objective： To establish an HPLC method for the determination of chlorogenic acid, caffeic acid and ferulic acid in Dabaidu capsules. Method： The column was an Agilent C 18 （250 mm× 4.6 mm,5 μm）column, the mobile phase was aeetonitrile- 0.4% phosphoric acid solution （15： 85）. The flow rate was 1.0 ml · min^-1 and the detection wavelength was 324 nm. Result： The linear range of chlorogenic acid was 5.42-32.43 μg · ml ^-1 （ r = 0.999 8 ） with the average recovery of 99.98% （ RSD = 0.2% , n = 6）. The linear range of caffeic acid was 2.63-26.32 μg · ml ^-1 （ r = 0. 999 4） with the average recovery of 99.31% （ RSD = 0.4%, n = 6）. The linear range of ferulic acid was 1.44-14.44 μg · ml ^-1 （ r = 0. 999 9） with the average recovery of 99.48% （RSD = 0.8% , n =6 ）. Conclusion： The method is simple, rapid and accurate with good reproducibility, and suitable for the determination of chloro- genic acid, caffeic acid and ferulic acid in Dabaide capsules.
