摘  要:采用1-苯基-3-甲基-5-吡唑啉酮(PMP)柱前衍生高效液相色谱(HPLC)法分析油茶多糖的单糖组成以及单糖摩尔比,确定油茶多糖单糖组成的分析条件。结果显示,油茶多糖由甘露糖、鼠李糖、葡萄糖醛酸、半乳糖醛酸、葡萄糖、半乳糖和阿拉伯糖7种单糖组成,摩尔比为:1.00:2.89:1.13:1.76:51.85:8.14:4.57,且其分子量大于一万。该法简单、准确、重现性好,可用于分析油茶多糖的单糖组成及摩尔比。Analysis of monosaccharide compositions and monosaccharide molar ratio in Camellia polysaccharides by 1-phenyl-3-methyl-5-pyrazolone (PMP) by pre-column derivatization high performance liquid chromatography (HPLC), and determine the Camellia polysaccharides monosaccharide compositions' analysis conditions. The results of HPLC analysis showed that Camellia polysaccharides consisted of Man, Rha, GlcUA, GalUA, GIc, Gal, Arab with the molar ratio of 1.00: 2.89:1.13:1.76:51.85:8.14:4.57, and the relative molecular mass is more than 10000. The experiment shows that HPLC method is simple, accurate and good repeatability, which can be applied to analyze the monosaccharide compositions and calculate the monosaccharide molar ratio in Camellia polysaccharides.
