摘 要:目的 检测通过颈动脉注射人骨髓间质细胞(hMSCs)移植趋向小鼠脑胶质瘤.方法 分离培养hMSCs,检测其CD73、CD105和CD90的表达;分析U87细胞条件培养基对体外hMSCs迁徙的作用,测定U87细胞培养液上清中转化生长因子(TGF)-β1和血管内皮生长因子(VEGF)的表达;通过颈动脉注射移植携带绿色荧光蛋白(GFP)的hMSCs,观察其向裸小鼠U87脑移植瘤模型趋向及肿瘤新生血管的生成情况.结果 分离培养的hMSCs呈长梭形、折光性强的成纤维样细胞形态,高表达CD73、CD105和CD90; U87条件培养基可以促进hMSCs的细胞迁移,在培养3d时,U87细胞培养上清的TGF-β1为(0.633±0.096) μg/L,而VEGF为(4.065±0.189) μg/L;将GFP标记的hMSCs通过颈动脉注射移植,3d后在裸小鼠脑内可观察到带有绿色荧光的细胞.结论 颈动脉注射移植的hMSCs能够趋向迁徙到裸小鼠脑胶质瘤,可能与肿瘤分泌的细胞因子相关.Objective To elucidate the distribution of human mesenchymal stem cells (hMSCs)homing to glioma after carotid injection.Methods hMSCs were isolated and maintained in MEM-α supplemented with 10% fetal bovine,and CD73,CD105 and CD90 expression was detected by flow cytometry.Intracranial gliomas xenografts mode was made by implanting U87 cells into the right frontal lobe of nude mouse.hMSCs labelled with green fluorescent protein (GFP) were injected into the right carotid artery of mice,and the homing of hMSCS toward gliomas was evaluated by checking the number of GFP cells in tumor area,also the tumor angiogenesis of the tumor was calculated by CD31 immuostaining.Results hMSCs were fusiform,fibroblast-like morphology,and flow cytometric analysis showed high level of CD73,CD105 and CD90 expression.In vitro,hMSCs could migrate in the U87 conditioned media.Also the amount of transforming growth factor-β1 (TGF-β1) and vascular endothelial growth factor (VEGF) in cultural supernatant of U87 cell was (0.633 ±0.096) and (4.065 ±0.189) μg/L after culture for three days.At three day following the injection of GFP labelled hMSCs,the GFP labeled cells were found distributed inside of tumor.Conclusion hMSCs by carotid injection could home to the intracranial gliomas xenografts mice mode,TGF-β1 may play a role in the attraction of hMSCs..
