摘  要:目的:研究慢性温和不可预知应激(CUMS)模型大鼠血浆中正五聚蛋白3(PTX-3)的质量浓度变化,及舍曲林对其的干预作用,寻找抑郁早期标志物。方法:取大鼠随机分为空白对照组、模型组和舍曲林(4.5 mg/kg)组,每组8只,♀♂各半,后2组大鼠每天给予应激刺激建立CUMS模型;于第11天开始舍曲林组大鼠灌胃给予相应药物,对照组和模型组大鼠给予等体积蒸馏水。分别于应激前、应激第7、11、18、21、24、27天考察各组大鼠的体质量、糖水消耗百分率、5 min内的行走和站立次数,以及血浆中PTX-3质量浓度。结果:与空白对照组比较,模型组大鼠应激第24天体质量、应激第21、27天糖水消耗百分率均明显降低(P<0.05),应激第11、18、21、24、27天行走次数和应激第18、21、24、27天站立次数均明显减少(P<0.05或P<0.01),应激第24天PTX-3质量浓度明显增加(P<0.05);与模型组比较,舍曲林组大鼠应激第27天糖水消耗百分率和行走次数、应激第21、24天站立次数均明显增加(P<0.05或P<0.01),应激第18、21、24、27天PTX-3质量浓度明显降低(P<0.05或P<0.01);其余各组间指标比较差异无统计学意义(P>0.05)。结论:PTX-3质量浓度随抑郁程度增加而增加,舍曲林干预后PTX-3质量浓度降低,提示PTX-3可作为抑郁早期标志物,但有待进一步研究论证。OBJECTIVE: To study the changes of plasma concentrations of Pentraxin-3 (PTX-3) in chronic unpredictable mild stress (CUMS) model rats and the intervention effects of sertraline against it in order to find out early diagnostic marker. METH- ODS: Rats were randomly divided into blank control group, model group and sertraline group (4.5 mg/kg) with 8 rats in each group (evenly divided between male and female). CUMS model was established in the latter 2 groups aiter daily stress stimulation. Sertraline group was given relevant medicines intragastrically since 11th day, and control group and model group were given con- stant volume of distilled water. Body weight, the percentage of sugar consumption, 5 min walking and standing times as well as plasma concentrations of PTX-3 were determined before stimulation, and 7th, 11th, 18th, 21st, 24th, 27th day of stimulation. RE- SULTS: Compared with blank control group, the body weight on 24th of stimulation, the percentage of sugar consumption on 21st and 27th of stimulation decreased significantly in model group (P〈0.05) ; walking times on llth, 18th, 21st, 24th and 27th of stimulation and standing times on 18th, 21st, 24th and 27th day of simulation decreased significantly (P〈0.05 or P〈0.01); plas- ma concentration of PTX-3 increased significantly on 24th day (P〈0.05). Compared with model group, the percentage of sugar consumption and walking time on 27th day of simulation, standing times on 21st and 24th of simulation increased significantly in sertraline group (P〈0.05 or P〈0.01); plasma concentrations of PTX-3 decreased significantly on 18th, 21st, 24th and 27th day (P〈0.05 or P〈0.01). There was no statistical significance in above index in the rest groups (P〉0.05). CONCLUSIONS: PTX-3 concentration increases with the increase of depression degree, while the concentration of sertraline decreases. Therefore, further study for PTX-3 as an early depression maker is needed.
