摘  要:为了获得最佳发酵培养基,提高凝结芽孢杆菌液体发酵水平,在单因素试验优化基础上,采用Plackett-Burman设计确定淀粉、大豆粉和鱼粉混合物(质量比2∶1)和磷酸氢二钾为凝结芽孢杆菌液体发酵水平的显著影响因素,通过最陡爬坡路径试验、中心组合设计和响应面分析法确定最优培养基组成。试验结果表明,最优培养基组成为淀粉10.6g/L、大豆粉13.5g/L、鱼粉6.7g/L、磷酸氢二钾2.72g/L、磷酸二氢钠0.312g/L、碳酸钙0.2g/L、一水硫酸锰0.169g/L、硝酸钠0.34g/L和七水硫酸镁0.74g/L。在最佳培养基组成条件下,液体发酵活菌数达67×108 CFU/mL。Researchs on fermentation medium of Bacillus coagulans strain applied to animal on singlefactor experiment basis have been done.Three important factors of starch,mix of soybean meal and fish meal(mass ratio was 2 to 1)and dipotassium hydrogen phosphate,which can influence the living bacteria amount,were screened through Plackett—Burman design and test.The results showed that the optimal medium was starch 10.6 g/L,mix of bean flour 13.5 g/L and fish meal 6.7 g/Land dipotassium hydrogen phosphate 2.72 g/L by using the steepest ascent path,center combination of design and response surface analysis.Under the optimized medium,the living bacteria amount of fermentation broth can reach 67 × 108CFU/mL.
