摘 要:目的通过研究β-连环蛋白(β-catenin)与胃癌组织分化程度及转移的关系,探讨β-catenin在胃癌发生及其转移的病理过程中的作用机理。方法应用Western Blot技术及免疫组化方法分析β-catenin在非恶性胃部组织、胃癌原发灶及转移灶组织中的表达,并对其进行比较。结果 Western Blot技术发现β-catenin在胃癌和非恶性胃部组织之间在表达的定量方面没有显著差异。免疫组化技术发现在163例原发性胃癌组织标本中,有79例β-catenin在细胞膜或者细胞浆阳性表达。β-catenin在低分化胃癌中表达水平显著降低(P<0.01)。41例转移性胃癌组织标本中有2例表达β-catenin,与原发性胃癌组织相比,表达水平显著降低(P<0.01)。结论β-catenin在部分原发性胃癌中表达缺失,在低分化胃癌及转移中表达缺失更为常见。Objective Through the study of β-catenin and gastric cancer tissue differentiation and metastasis, to study the role of β-catenin in the pathogenesis of gastric cancer and its metastasis. Methods To comparison the expression of β-catenin in nonmalignant stomach tissues, in human gastric cancers and metastasis tissues,by using immunohistochemistry and Western blot analysis. Results There were no quantitative differences between gastric cancers and the nonmalignant gastric tissues, as determined by Western blot analysis. Immunohistochemistry analysis showed that β-catenin expression was present at either the cell membrane or the cytoplasm in 79 of 163 primary gastric cancers.β-catenin in poorly differentiated gastric cancer group decreased significantly(P〈0.01). Two of 41 metastatic cancer lesions expressed β-catenin protein.Expression level significantly reduced(P〈0.01) with primary gastric cancer tissues compared. Conclusion β-catenin expression is lost in a subgroup of primary gastric cancers, is frequently absent in poorly differentiated gastric cancer group and metastases.
