摘 要:目的探究不同含量反式脂肪酸(trans fatty acid,TFA)饮食对小鼠肝脏氧化应激及PPARγ表达的影响及可能机制。方法成年雄性C57BL/6小鼠40只,随机分为空白对照(CN)、低(L-TFA)、中(M-TFA)、高剂量(H-TFA)反式脂肪酸饮食4组,每组10只。饲喂处理90 d后,分别测量各组动物的净增体重和食物转化率;全自动生化分析仪检测血清甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白(HDL-C)、低密度脂蛋白(LDL-C)、游离脂肪酸(FFA)水平;化学比色法测定肝组织中丙二醛(MDA)含量及谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT),碱性磷酸酶(ALP)、丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(AST)、乳酸脱氢酶(LDH)免疫组化分析肝组织PPARγ阳性表达变化。结果与CN组比较,M-TFA和H-TFA组小鼠净增体重和食物转化率显著增加(P<0.05);TG、TC、LDL-C、FFA等血脂水平显著增加(P<0.05);肝组织中MDA含量显著增加(P<0.05),GSH-Px、SOD、CAT、活性显著降低(P<0.05)ALP、ALT、AST、LDH活性显著升高(P<0.05);光镜下观察反式脂肪酸饮食小鼠肝组织均见一定程度的脂质沉积,胞浆内充满脂肪空泡或脂滴;PPARγ免疫阳性表达显著增强(P<0.05),且阳性染色主要定位于脂质沉积的肝实质细胞核内。结论中、高剂量反式脂肪酸饮食可诱导肝氧化应激损伤,脂质沉积,PPARγ蛋白表达上调。Objective To investigate the effects of diet with different trans fatty acid (TFA) levels on oxidative stress in liver and expression of PPAR'~ in mice. Methods Forty adult C57BL/6 mice were randomly divided into four groups (n= 10): Control (CN), L-TFA (25 mg/kg'bw), M-TFA (50 mg/kg.bw), H-TFA (100 mg/kg'bw). All mice were fed different diets for 90 d. The body weight gain, feed efficiency, and the serum levels of TG, TC, HDL-C, LDL-C, FFA were determined. The automatic biochemical analyzer was used for blood analysis. The spectrophotometry was used to determine the activities of SOD, GSH-Px, CAT, ALP, ALT, AST, LDH and the content of MDA in liver. At the same time, the number of PPAR7 positive cells was estimated by immunohistochemistry. Results Compared with CN, body weight gain and feed efficiency were significantly increased in M-TFA and H-TFA group. The serum levels ofTG, TC, LDL-C, FFA significantly, and the MDA levels and ALP, ALT, AST, LDH activities in liver were significantly increased, but the SOD, GSH-Px activities CAT significantly decreased. The liver damage was displayed in moderate to severe cytoplasm vacuolation, hepatocyte hypertrophy, hepatocyte ballooning and necroinflammation. The number of PPARy positive cells was increased in M-TFA and H-TFA groups. Conclusion Higher TFA levels may induce oxidative stress, lipid deposition and up expresssion of PPARy in liver.
