摘 要:目的探讨睫状神经营养因子(CNTF)及其受体(CNTFRа)在脊髓损伤中的变化及亚硒酸钠对脊髓损伤的保护作用。方法将不同年龄大鼠(幼年、成年、老年)随机分为对照组、模型组及亚硒酸钠处理组,阻断腹主动脉建立脊髓损伤模型,对照组不夹闭血管,亚硒酸钠处理组于术前2 w开始补充营养剂量的亚硒酸钠并夹闭腹主动建立脊髓损伤模型。分别于伤后1d、7d、14d处死动物,取脊髓分别作HE染色、免疫组化及RT-PCR检测,观察神经细胞结构及CNTF、CNTFRа的变化。结果正常大鼠各年龄组均有CNTF、CNTFRа蛋白及mRNA表达,幼年组强于成年组及老年组(P<0.05);亚硒酸钠干预组CNTF mRNA、CNTFRa mRNA表达较模型组增强,增强持续时间幼年组最长。结论睫状神经营养因子对大鼠脊髓损伤有保护作用,亚硒酸钠能上调CNTF、CNTFRа的表达。Objective To investigate the alteration of ciliary neurotrophic factor (CNTF) and its receptor (CNTFRa) after spinal cord injury and the protective effect of sodium selenite. Methods The different-aged rats were randomly divided into control group, experimental group and sodium selenite treated group. In experimental group, the abdominal aorta were ligated; In sodium selenite treated group, the rats were fed nutritional dosage of sodium selenite before ligating their abdominal aorta. After 1 d, 7 d or 14 d, the rats were killed to investigate the structure of nerver cells by HE staining and the expression of CNTF and CNTFRa by immunohistochemical method and RT-PCR. Results Both protein and mRNA of CNTF and CNTFRa were positively expressed in different-aged rats of control group. The mRNA levels of CNTF and CNTFRa in sodium selenite treated group were higher than experimental group. Conclusion Sodium selenite can up-regulate the expression of CNTF and CNTFRa. Ciliary neurotrophic factor plays protective roles agaisat the development of spinal cord injury.
