摘 要:为了获得沙苑子黄酮类化合物的有效提取工艺参数,在单因素试验基础上选取液料比、提取温度、沙苑子粒度、提取时间作为试验因素,以提取物的吸光度为响应值,采用响应面法对提取工艺参数进行优化,并对优化条件下的提取物进行清除DPPH自由基能力研究。结果表明:60%甲醇是有效的提取剂;最佳工艺参数为粒度120目,温度42℃,提取时间47 min,液料比30∶1,该条件下沙苑子黄酮类物质的粗提率为10.66%;提取物具有一定的清除DPPH自由基效果,低浓度条件下呈现良好的量效关系,浓度为2%时清除率最高。In order to obtain the optimum extraction conditions for flavoness from Semen Astragali Complanati,on the basis of one-factor experiment,the ratio of solvent-to-sample,extraction temperature,particle size and extraction time were optimized by Response Surface Methodology (RSM) and the absorption of extracts was chosen as the response value.The optimized extraction conditions were determined as follows:the suitable extractant was 60% methanol;the particle size was 120 mesh;the extraction temperature was 42 ℃ ;the extraction time was 47 min;the ratio of solvent-to-sample was 30:1.Under these conditions,the crude extraction yield of flavone was 10.66%.In addition,the DPPH scavenging ability of the extract was investigated.The results indicated that there was a good correlation between extract's concentration and scavenging rate at lower concentrations,and the highest DPPH scavenging ability was obtained when the concentration of flavone was 2%.
