明胶酶和Twist在乳腺癌组织及患者血清中的表达及临床意义

摘　　要：目的观察乳腺癌及乳腺良性病变患者血清明胶酶[基质金属蛋白酶-2(matrix metalloproteinases-2,MMP-2)、MMP-9]和Twist的水平,探讨其与组织中表达的关系。方法采用免疫组化MaxVision两步法和ELISA法检测46例乳腺癌和31例乳腺良性病变组织中MMP-2、MMP-9和Twist蛋白的表达及患者术前血清MMP-2、MMP-9和Twist水平。结果 MMP-2、MMP-9和Twist蛋白和术前血清水平在乳腺癌组均明显高于乳腺良性病变组(P<0.05);乳腺癌组织MMP-2、MMP-9和Twist蛋白阳性率均随着TNM分期的增高而增高,且在腋淋巴结转移组均高于无转移组(P<0.05);乳腺癌患者术前血清MMP-2、MMP-9和Twist水平与癌组织相应蛋白表达均呈显著正相关(P<0.05)。结论血清中MMP-2、MMP-9和Twist水平改变可以反映组织中其表达水平,其检测对乳腺癌的转移预警和辅助诊断治疗具有一定的临床指导意义。Purpose To investigate the level of MMP-2, MMP-9 and Twist in serum of breast carcinoma and to study its relationship with their expression in tumor tissues. Methods The level of MMP-2, MMP-9 and Twist in serums and tissues of 46 cases of breast carcinoma and 31 cases of benign breast lesions were analyzed by immunohistochemical staining and enzyme-linked immunosorbent as- say. Results MMP-2, MMP-9 and Twist protein and serum levels in breast cancer group were significantly higher than those in benign breast lesions （ P 〈 0.05 ）. Increased expression of MMP-2, MMP-9 and Twist protein was associated with high TNM stage and tumor lymph node metastasis （ P 〈 0.05 ）. Significant positive correlations were observed between serum and tissue levels of MMP-2, MMP-9 and Twist protein, respectively （ P 〈 0. 05 ）. Conclusion The serum levels of MMP-2, MMP-9 and Twist were correlated with their tissue expression, suggesting the detecting of serum levels would be useful for predicting metastasis and early warning and clinical diag- nosis and therapy.
