重组sRAGE对肝衰竭模型鼠肝组织TM表达的影响

摘  要:[目的]探索可溶性晚期糖化终末产物受体(receptor for advanced glycation end products,RAGE)对肝衰竭模型小鼠肝组织表达血栓调节蛋白(thrombomodulin,TM)的影响。[方法]原核表达纯化得到rsRAGE,尾静脉注射rsRAGE至急性肝衰竭模型小鼠,用不同方法检测肝组织TM表达、TM-mRNA相对空白组变化趋势、血清中谷丙转氨酶(ALT)和可溶性TM(sTM)的含量。[结果]成功构建PET-28a-rRAGE原核质粒,表达并纯化出分子量大约为31600的蛋白。实验组24h生存率为75.0%明显高于对照组的45.8%;苏木精-伊红染色显示实验组肝组织损伤程度较对照组减轻;免疫组化检测实验组TM24h积分吸光度(I A)值为25.70±5.96显著小于对照组(70.24±5.65),P<0.05;RT-PCR检测实验组TM-mRNA表达为41.23±5.20,峰值显著低于对照组(75.26±13.66),P<0.05;实验组血清ALT峰值为(68.33±10.29)U/L明显低于对照组[(114.23±13.59)U/L],且峰值出现推后;实验组血清sTM峰值为(4.63±9.44)ng/ml明显低于对照组[(9.91±1.08)ng/ml],P<0.05。[结论]rsRAGE可以显著减少肝衰竭模型小鼠肝组织TM的表达,降低血清sTM的含量,降低血管内皮损伤程度,且降低血清谷丙转氨酶含量,改善肝功能,减小肝组织损伤。[Objective]To study the influence of soluble advanced glycation end products receptor on the expression of thrombomodulin(TM) in liver failure mouse model. [Methods]Acute hepatic failure was in- duced by D-galactosamine and lipopolysaccharide in mice. After injection with saline or rsRAGE proteins, the expression of TM in liver tissues was detected by immunohistochemisty and RT-PCR, and the serum ALT and sTM were quantified. [Results]The recombinant prokaryotic expression vector PET-28a-rRAGE was successfully constructed and mouse sRAGE protein about 31600 was successfully expressed. RsRAGE alleviated the acute liver damage. RsRAGE treatment decreased the expression of TM in liver tissues and reduced the serum levels of ALT and sTM. [Conclusion]RsRAGE can alleviate the acute liver damage and inhibit the expression of TM.
