摘 要:目的 探讨高胆固醇膳食诱导胆囊胆固醇结石形成小鼠中肝细胞小窝蛋白-1(Cav-1)的表达及其与胆囊胆固醇结石形成的相关性及其作用机制.方法 C57L小鼠24只,随机均分为对照组、致石组,分别给予普通膳食与高胆固醇膳食,饲养8周建立胆囊胆固醇结石模型,检测并比较两组小鼠血清生化指标,并利用逆转录-聚合酶链反应(RT-PCR)与Western blot方法检测小鼠肝细胞Cav-1在mRNA与蛋白水平的表达.结果 致石组小鼠全部成石.致石组小鼠的血清胆固醇[(4.19±1.02) mmol/L]、HDL[(2.12 ±0.27) mmol/L]、LDL[(2.67±1.28) mmol/L]、总胆汁酸[(75.79±78.25) mmol/L]含量均较对照组增高;致石组小鼠肝细胞内Cav-1在mRNA水平(2.28±0.05;t=32.714,P<0.01)与蛋白水平(1.68±0.08;t=2.886,P<0.05)的相对表达量均较对照组增高.结论 高胆固醇膳食诱导小鼠胆囊胆固醇结石形成过程中,肝细胞内Cav-1在短期内表达水平增高,但Cav-1的表达改变是否与胆固醇结石形成相关,尚不确定.Objective To investigate the Caveolin-1 (Cav-1) expression in hepatocytes in mice with high cholesterol diet-induced cholesterol gallstone and its correlation with cholesterol gallstone forma tion and the possible mechanism. Methods Twenty-four C57L mice were randomly divided into control group and gallstone group (n = 12 each), which were fed on normal diet and high cholesterol diet separate ly for 8 weeks. Then measured and compared two groups of mice Serum biochemical indicators, and the ex pression of Cav-1 mRNA and protein were measured in hepatocytes of mice. Results In gallstone group cholesterol gallstone model was successfully established in all mice. The serum cholesterol E (4. 19 ± 1.02) mmol/L], HDL [ (2. 12 ±0. 27) mmol/L], LDL [ (2.67 ± 1.28) mmol/L, and total bile acid content [ (75.79 ±78.25) mmol/L] in gallstone group were higher than in control group. The Car-1 relative ex pression (2. 28 ±0. 05 for mRNA;t =32. 714,P 〈0. 01 ; 1.68 ±0. 08 for protein,t=-2. 886,P 〈0.05) in gallstone group was higher than in control group. Conclusion During the gallstone formation process in the mice with high cholesterol diet-induced cholesterol gallstone, the Car-1 expression in hepatoeytes is in creased in short-term, buth whether the change of expression of Cav-1 is associated with cholesterol gall- stone formation is uncertain.
