摘 要:目的 分离并鉴定成石与未成石胆汁内代谢物,推测其可能的分子式及结构.方法 健康豚鼠30只,致石饲料饲养8周,采集胆囊胆汁、胆石标本,高效液相色谱分离胆汁,并行质谱分析.结果 成石胆汁中质荷比(M/Z)质量数为483.2、515.8、484.2、312.7 amu的物质含量明显升高,质量数为277.9、295.0、297.1、506.2、449.0 amu的物质含量明显降低.结论 饮食诱导豚鼠胆囊胆色素结石形成过程中,胆汁中牛磺胆酸、牛磺石胆酸、硬脂酸等的含量增高,甘氨胆酸及亚麻酸等含量下降.液质联用进行胆汁代谢组学研究可行.Objective To isolate and identify bile metabolites in guinea pigs with stone formation and speculate their possible molecular formula and structure. Methods Pigment gallstone-forming forage was given to 30 healthy guinea pigs for 8 weeks. The bile metabolomics was detected by using liquid chro matography-mass spectrometry (LC-MS). Results Compared with the gallbladder bile without stone, me tabolites with M/Z for the quality of 483.2, 515.8, 484. 2 and 312. 7 ainu were significantly increased, and those with M/Z mass number for 277. 9,295.0,297. 1,506. 2 and 449. 0 ainu were decreased signifi cantly. Conclusion During the process of diet-induced pigment gallstone formation in guinea pigs, the changes of bile composition occurred. The levels of taurocholic acid and stearic acid were significantly in- creased, and those of glycocholic acid and linolenic acid are decreased. It was feasible for bile metabolo mics stud' with LC-MS.
