摘 要:通过以油烟冷凝液为惟一碳源的选择培养基初筛,从长期受到油烟严重污染的土壤中筛选出具有降解油烟污染物能力的优势菌株XJ01、XJ03。研究了其最佳生长条件及降解性能,并对其进行了生理生化特征及分子生物学鉴定。研究表明,XJ01、XJ03最佳生长温度分别为35℃、30℃,最佳生长pH均为7,摇床转速均为120 r/min,最佳降解时间分别为48 h和36 h;在最佳降解条件下,XJ01、XJ03总降解量分别达4.47 mg/L和4.18 mg/L,降解过程中,降解液pH值持续下降,生物量先增加后下降。经分子生物学鉴定,菌株XJ01、XJ03分别与铜绿假单胞菌群(Pseudomonas aeruginosa group)和克雷伯氏菌属(Klebsiella sp.)同源性最高均达到100%。Two strains named as XJ01 and XJ03 which could degrade pollutants from cooking fume effi- ciently were isolated from soils that were seriously polluted by cooking fume for long time, and were screened on the plates in the selective medium with fume condensate as sole carbon source. Experiments were conducted to study the growth and degradation conditions of the two predominant strains, meanwhile, the identification on morphological, physiological-biochemical property and molecular biology were studied. The results showed that the optimum growth temperatures of XJ01, XJ03 were 35 ℃ and 30℃ and the optimum degradation times were 48 h and 36 h, respectively. The optimum growth pH and rotation speed was 7 and 120 r/rain for two strains. Un- der the optimum degradation conditions, strain XJ01, XJ03 was with the total degradation quantity of pollutants from cooking fume as high as 4. 47 mg/L and 4. 18 mg/L, respectively. Along with the increase of degradation time, the pH value of degradation liquid continously declined, biomass increased first and then decreased. The result of molecular biology identification showed that strain XJ01, XJ03 shared 100% homologies with Pseudo- monas aeruginosa group and Klebsiella sp. respectively.
