硫酸软骨素对Lewis肺癌细胞增殖及侵袭能力的影响

摘　　要：目的观察硫酸软骨素(CS)对Lewis肺癌细胞(LLC)增殖及侵袭能力的影响,并探讨其可能机制。方法常规方法培养LLC,并给予含不同剂量CS的培养液培养LLC。采用MTT法检测CS对LLC增殖的影响,流式细胞术PI染色法检测CS对LLC细胞周期的影响,Transwell侵袭实验检测CS对LLC侵袭能力的影响。结果与对照组比较,CS组A值随CS浓度增加而降低(P均<0.05);与对照组比较,9、30 mg/mL CS组S期升高(P均<0.05);与对照组比较,9、30 mg/mL CS组LLC穿膜细胞数降低(P均<0.05)。结论 CS对Lewis小鼠肺癌细胞具有抑制增殖和侵袭的作用,其机制可能与抑制细胞分裂周期有关。Objective To investigate the effects and mechanism of chondroitin sulfate （CS） on cell proliferation and invasion of mouse Lewis lung carcinoma cells （LLC）.Methods LLC were cultured by routine method and treated with CS at different concentrations.The effects of CS on LLC proliferation were examined by MTT.The cell cycle of LLC treated with CS was observed by FCM via PI staining.The invasion of LLC treated with CS was detected by Transwell assay.Results Compared with the control group,the proportion of G0/G1 and G2/M was reduced significantly with the increased concentrations of CS （all P ＜ 0.05）,the proportion of S was increased significantly after being treated with 9 and 30 mg/mL CS in the CS group （all P ＜0.05）.Meanwhile,the number of cell-permeating septum in the CS group was significant less than that of the control group （all P ＜ 0.05）.Conclusion CS suppresses the proliferation and invasion of LLC with the possible mechanism of blocking the cell cycle.
