摘  要:目的通过分析抑郁模型大鼠脑组织谷氨酸(Glu)和γ-氨基丁酸(GABA)浓度的变化,探讨抑郁症发病的神经化学机制。方法 54只雄性SD大鼠随机分成2批,每批分为正常对照组、模型组和氟西汀组。正常对照组合笼饲养,其余各组单笼孤养。模型组和氟西汀组建立慢性轻度不可预见性应激(CUMS)大鼠模型。氧西汀组大鼠于造模开始即接受0.5%羧甲基纤维素钠(CMC)悬浮液配制的氟西汀[1.54 mg/(kg·d)]灌服21 d,正常对照组和模型组大鼠灌服0.5%CMC悬浮液21 d,1次/d。采用CUMS结合孤养建立抑郁大鼠模型,第1批30只大鼠用于自发活动、糖水消耗和强迫游泳测试;第2批24只大鼠用于库仑阵列-电化学高效液相色谱法测定内侧前额叶皮质(mPFC)和海马Glu及GABA的浓度。结果与正常对照组或氟西汀组比较,模型组大鼠自发活动显著减少(P<0.01),糖水消耗量及糖水偏爱百分比明显降低(均P<0.05),强迫游泳不动时间延长(P<0.01);mPFC和海马Glu浓度均明显升高(P<0.05,P<0.01),GABA浓度明显降低(P<0.05,P<0.01)。氟西汀组与正常对照组比较,上述各项指标差异均无统计学意义(P>0.05)。结论 CUMS抑郁模型大鼠mPFC和海马存在Glu和GABA递质系统功能紊乱,氟西汀治疗可改善上述神经递质异常。Objective To evaluate the concentration change of glutamate (Glu) and gamma -aminobutyric acid (GABA) in medial prefrontal cortex (mPFC) and hippocampus in chronic unpredictable mild stress (CUMS) -induced depression rats, thus to investigate the related neurobiochcmistry of depression. Methods Fifty - four male adult SD rats were randomly divided into two groups, moreover, each group was subdivided into three subgroups: control group, model group and fluoxetine -treated group. The depression rat model was established under CUMS and separated feeding, after which, an open field, sugar consumption and forced swimming tests were applied in the first group of 30 rats. The brains tissues of the second group were collected for analysis of the levels of Giu and GABA in mPFC and hippocampus coularray electrochemic detection for high performance liquid chromatogram technique. Results Compared with both the control group and the fluoxetine - treated group, significant reduction in spontaneous activity, sucrose consumption, preference percentage and immobility time in forced swimming was observed in model group, with significant up - regulation of Glu and down - regulation of GABA in mPFC and hippocampus ( P 〈 0. 05 ). There was no significant difference between control group and fluoxetine - treated group. Conclusion The activity of Glu and GABA systems in the mPFC and hippocampus is abnormal in CUMS -induced depression rats, which is amended with fluoxetine treatment.
