摘 要:目的测定新疆不同地区和品种的葡萄树伤流液中氨基酸含量,为葡萄树伤流液药用、食用价值提供理论依据。方法采自新疆和田市、吐鲁番鄯善县、喀什市和阿图什市的9种葡萄树伤流液,采用异硫氰酸苯酯(PITC)为柱前衍生化试剂衍生化后进行高效液相色谱(HPLC)分析,建立葡萄树伤流液中17种氨基酸含量测定方法,并对其特征氨基酸进行评价。结果9个品种的葡萄树伤流液中氨基酸有17种,总含量为6.20~201.22mg/L,其中7种必需氨基酸。主成分分析得出组氨酸、酪氨酸、缬氨酸、异亮氨酸、苯丙氨酸、天冬氨酸和丙氨酸是葡萄树伤流液的特征氨基酸。结论不同品种葡萄树伤流液的氨基酸含量差异较大,但是各类氨基酸含量分布与组成类似。主成分分析法是葡萄树伤流液氨基酸含量分析的有效方法。Objective To determine the amino acid content of different kinds bleeding sap of grape tree from different areas of Xinjiang, and provide a theoretical basis on medicinal and food value of bleeding sap of grape trees in Xinjiang. Methods Derivated by phenylisothiocyanate (PITC), the determination on 17 kinds of amino acids in 9 grapevine bleeding sap was established, which were collected from Hotan, Tur- pan and Atushi. The characteristic amino acids were evaluated. Results There were 16 kinds of amino acids in bleeding sap of Xinjiang grape tree and their contents were 6. 204201.22 mg/L, and 7 of them were essential for human body. The analysis results of principal components showed that histidine, tyro- sine, valine, isoleucine, phenylalanine, asparticacid may be the characteristic amino acids. Conclusions There was significant difference between contents of various amino acids, but their distribution and compo sition were similar. Principal component analysis is an effective way to analyze the amino acid contents in bleeding sap of grape trees.
