摘  要:目的建立初断乳雄性SD大鼠铅损伤动物模型,用于研究铅对动物主要器官组织系统的损害。方法选取45只初断乳雄性SD大鼠,分层随机分成正常对照组、1‰和4‰浓度乙酸铅水剂量组,以自由饮用方式进行染毒28 d,实验结束时处死动物,进行各种生理指标及肝、脑、股骨铅含量检测。结果与饮用蒸馏水组相比,随着乙酸铅浓度的增加,雄性SD大鼠各项生理指标都反映出铅损伤迹象,肝、脑及股骨中的铅含量都呈升高趋势,饮用1‰和4‰乙酸铅水两个剂量组大鼠肝脏和股骨中铅含量有显著性升高(F=37.555,P﹤0.01;F=36.67,P﹤0.01),饮用4‰乙酸铅水剂量组大鼠脑中铅含量也有显著性升高(F=26.363,P﹤0.01)结论初断乳雄性SD大鼠饮用含乙酸铅蒸馏水建立动物铅损伤模型是一种简便、可靠、易于控制的方法,可用于对大鼠铅损伤的研究。OBJECTIVE The study aimed to develop an animal model for lead-induced damage using male weanling SD rats, so to study the damaging effect of lead on major organ systems of animalSl METHODS 45 male weanling SD rats were selected and randomly divided into a regular control group, a 1%o lead acetate group, and a 4%0 lead acetate group. The rats were exposed to lead by consuming lead acetate-containing water for 28 consecutive days, and were sacrified at the end of the experiment. Various physiological parameters and lead contents in liver, brain, and thighbone of the rats were measured. RESULTS As compared with the distilled water-consuming control group, physiological parameters of the rats in the expo- sure groups indicated signs of lead damage, and lead contents in liver, brain, and thighbone of the exposure groups in- creased with the increase in lead acetate concentration. Lead contents in liver and thighbone of the rats in the 1%o and 4%0 lead acetate groups significantly elevated (F = 37.555, P 〈 0.01 ; F = 36.67, P 〈 0.01) ; lead content in brains of the the 4%0 lead acetate group also increased significantly (F = 26.363, P 〈 0.01 ). CONCLUSION Establishing an animal model for lead-induced damage by feeding male weanling SD rats lead acetate-containing distilled water is a simple, reliable, and easily controllable method, which may be adopted in the study of lead-induced damage in rats.
