摘 要:目的了解快速眼动睡眠剥夺改善抑郁情绪的可能机制。方法将大鼠分为正常对照组(A组)、抑郁模型组(B组)、抑郁模型+72 h睡眠剥夺组(C组),在建立慢性轻度不可预见性应激的抑郁模型后,采用小平台水环境法对大鼠进行72 h快眼动睡眠剥夺,以强迫游泳实验检测大鼠的不动时间,下丘脑5-HT、腺苷浓度分别用高效液相色谱-荧光检测法和高效液相色谱-紫外检测法进行测定。结果 21 d慢性轻度不可预见性应激后,B组和C组大鼠的强迫游泳不动时间显著延长(P<0.05),快眼动睡眠剥夺后C组大鼠强迫游泳不动时间显著缩短(P<0.01);B组大鼠下丘脑5-HT浓度显著低于A组(P<0.01),C组大鼠下丘脑5-HT浓度显著高于B组(P<0.01);B组大鼠下丘脑腺苷浓度显著低于A组(P<0.05),C组大鼠下丘脑腺苷浓度显著低于B组(P<0.01)。结论快眼动睡眠剥夺可以逆转大鼠的抑郁样行为,并增加大鼠下丘脑5-HT浓度,其可能在睡眠剥夺抗抑郁过程中发挥了重要作用,腺苷可能参与了睡眠剥夺的抗抑郁过程。Objective To explore the underlying antidepressive mechanisms of rapid eye movement (REM) sleep deprivation.Methods 30 Sprague-Dawley rats were randomized into normal control group (A group,10 cases),depression model group (B group,10 cases),depression model with sleep deprivation group (C group,10 cases).Depression model was established by means of chronic unpredictable mild stress (CUMS) and 72 hours REM sleep deprivation was performed by means of flower pot technique.All the 30 rats were assessed by forced swimming test to record their motionless time.Level of 5-HT and adenosine in hypothalamus were measured by means of high-performance liquid chromatography (HPLC)-fluorescence detection and HPLC-ultraviolet detection respectively.Results Motionless time of forced swimming test increased significantly in rats in B group and C group after 21 days CUMS (P 〈 0.05),and it decreased significantly in rats in C group after 72 hours REM sleep deprivation (P 〈 0.01).Level of 5-HT in hypothalamus was significantly lower in B group than that in A group (P 〈 0.01),and it was significantly higher in C group than that in B group (P 〈0.01).Level of adenosine in hypothalamus was significantly lower in B group than that in A group (P 〈0.01),and it was significantly lower in C group than that in B group (P 〈0.01).Conclusion REM sleep deprivation can reverse the depression-like behavior of rats.It can also increase the concentration of 5-HT in hypothalamus,which may play an important role in the mechanism of antidepressant effect.The change of adenosine in hypothalamus may also contribute to the antidepressant process of REM sleep deprivation.
