膀胱移行细胞癌E-钙粘附蛋白和α连接素表达的定量研究

摘　　要：目的 :定量地检测E 钙粘附蛋白 (E cad)及α 连接素 (α CTN)在膀胱移行细胞癌的表达 ,并探讨它们与肿瘤生物学行为的关系。方法 :应用流式细胞术 ,检测 30例膀胱癌冰冻组织的E cad和α CTN的表达量。结果 :肿瘤细胞E cad(0 .5 6 77± 0 .3194) ,α CTN(0 .5 72 6± 0 .2 895 )的表达量 (荧光指数 ,FI)均明显低于正常粘膜(P <0 .0 1)。浸润型肿瘤二者表达量低于表浅型肿瘤 (P <0 .0 1) ,复发者E cad表达量低于初发者 (P <0 .0 5 )。E cad和α CTN表达量呈正相关 (P <0 .0 1)。结论 :膀胱移行细胞癌E cad和α CTN表达均下调 ,并以浸润型肿瘤为显著 ,肿瘤复发后 ,其E cad表达量降低。E cad和α CTN表达量呈正相关。二者下调可能具有因果关系。Objective: To measure the expression of E cadherin (E cad) and α catenin (CTN) in transitional cell carcinomas (TCC) of bladder and to study the relationship between the expression and biological behavior of the carcinoma. Methods:Expression of E cad and α CTN were measured by flow cytometry in 30 cases of frozen bladder carcinomas. Results: The fluorescence index (FI) of E cad and α CTN of tumor cells were 0.5677±0.3194 and 0.5726±0.2895, respectively,which were significantly lower than those of normal bladder mucosa ( P <0.01). Both were lower in invasive tumors than in superficial tumors ( P <0.01). E cad expression decreased in recurrent tumors ( P <0.05),and there was a positive correlation between the expressions of E cad and α CTN ( P <0.01). Conclusi on: The expression of E cad and α CTN were down regulated in TCC of bladder, and those of invasive tumors were lower than superficial tumors. E cad expression decreased when the tumor had recrudesced. E cad and α CTN have a positive correlation, which means they have some relationship of sequence.
