摘  要:鼠李糖乳杆菌GG是国家认可的第3代益生菌的一种,因其可在肠道定植并且有耐酸碱等特性而得到广泛应用。实验希望优化其培养基,使鼠李糖乳杆菌GG能用较低的成本来进行高密度培养。首先以MRS培养基为基础,根据物种特性进行碳源(麦芽糖:葡萄糖:乳糖:蔗糖)、氮源(胰蛋白胨:牛肉膏:酵母粉)和增菌物质(黄瓜汁:西红柿汁)的成分分别调整,之后进行单因素分析。在相同的条件下,经过37℃厌氧、恒温培养16 h后,测OD600来比较增菌效果。实验发现胰蛋白胨、酵母粉、葡萄糖、西红柿汁这4种物质增菌明显。在以上述4种物质设计4因素3水平L9(34)的正交分析,结果为鼠李糖乳杆菌GG影响性西红柿汁>胰蛋白胨>葡萄糖>酵母粉,其中最佳配比方案为A3B1C3D2(2.5%胰蛋白胨+1.5%酵母粉+2.5%葡萄糖+20%西红柿汁)。在优化配方下鼠李糖乳杆菌GG可达9.4×109cfu/mL。Lactobacillus rhamnosus GG was considered as a probiotic by the Chinese government. Because it can live in the intestinal colonization and survive through acid and alkali. It is more widely used. Therefore, we want to optimize the medium, so that the Lactobacillus rhamnosus GG can be producted at a lower cost of the cultivation of high-density. We use MRS medium as basement and respectively adjust the carbon source (maltose:glucose:lactose:sucrose), nitrogen source (tryptone:beef extract:yeast powder) and other ingredients (cucumber juice:tomato juice) to the univariate analysis, Under the same conditions, 37 %" anaerobic, thermostat culture after 16 h, measured the OD6oo tocompare the mount off cell. Experiments found that obvious enrichment effect of tryptone, yeast extract, glucose, tomato juice. Then we use the four substances four factors and three levels of L9(34) orthogonal analysis, the results of Lactobacillus rhamnosus GG impact was tomato juice〉tryptone〉glucose^yeast powder. So the best recipe was A3B1C3D2 (2.5% tryptone+1.5% yeast extract+2.5% glucose+20% tomato juice).
