摘 要:目的对脂肪细胞大小与血清胰岛素、瘦素水平的关系进行初步探讨。方法用高脂饲料喂养健康C57BL/6乳鼠建立肥胖小鼠动物模型,测量体重(Wt)、空腹血糖(FPG)、三酰甘油(TG)、血清胰岛素(FINS)及瘦素(LEP)水平,计算Lee’s指数及稳态模型胰岛素抵抗指数(HOMA-IR),脂肪组织HE染色观察脂肪细胞大小改变,统计学分析其相关性。结果与同周龄对照组小鼠相比,肥胖组小鼠Wt、FPG、TG、FINS、LEP和Lee’s指数及HOMA-IR均升高;相同放大倍数视野下,肥胖组小鼠脂肪细胞数目较正常组减少,脂肪细胞面积增大;统计学分析显示脂肪细胞大小与体重、血清胰岛素、瘦素和胰岛素抵抗指数呈正相关。结论肥胖小鼠脂肪细胞显著增大,易并发瘦素和胰岛素抵抗,且胰岛素抵抗严重程度与脂肪细胞大小呈高度正相关。[ Objective ] To investigate the correlation of adipocyte size, serum insulin and leptin levels in high-fat induced obesity mouse. [Methods ] Obesity mouse model was established through feeding healthy C57BL/6 mice with high-fat diet. The weight, blood glucose, blood lipid, serum insulin, serum leptin, Lee's index and HOMA-IR were assayed or calculated. After adipose tissues stained with HE, the adipocyte size was quantified and the correla- tion was analyzed. [Result] The weight, blood glucose, blood lipid, serum insulin, serum leptin, HOMA-IR and adipoeyte size in obesity group were significantly higher than those in normal group. And the statistical analysis sug- gests that the adipoeyte size of high-fat-induced obesity mouse is positively correlated to its weight, serum insulin, serum leptin levels and HOMA-IR. [ Conclusion] The obesity mouse with increased adipoeyte size is more easily complicated by leptin and insulin resistance. Moreover, the severity of insulin resistance is highly correlated to adipocyte size.
