摘 要:目的 研究旁观者效应在TK/GCV系统杀灭肿瘤细胞中的作用。方法 转TK基因肺癌细胞A549-TK与亲代细胞A549分别以不同比例、高低密度混合培养,加50 (mol/L GCV作用5 d,MTT法测活。结果 当细胞以高密度接种(104/孔),TK+细胞占20%就能观察到较对照细胞出现明显的生长抑制(P<0.05),而细胞以低密度接种(103/孔),TK+细胞占50%出现较对照细胞明显地生长抑制 (P<0.05)。结论 旁观者效应在TK/GCV系统杀灭瘤细胞中起重要作用,且这一效应与细胞间密切接触有关。Objective To study the role of bystander effect in the cytocidal activity of TK/GCV against tumor cells. Methods Human pulmonary carcinomal cell line and TK gene-transfected A459 cells were cultured together in varied ratio at low and high cell density respectively, treated with 50 mol/L GCV for 5 d. The living cells were counted by MTT assay. Results When the cells were cultured at a high density (104/well) with only 20% TK+cells, significantly higher growth inhibition than that of control cells (P<0.05) was observed. Obvious growth inhibition was observed (P<0.05) in the cell culture at a low density (103/well) only when TK+cell ratio was raised to 50%. Conclusion Bystander effect plays an important role in the cytocidal activity of TK/GCV against tumor cells, and is quite relative to intercellular contact.
