摘 要:目的:考察复方枳楔子制剂(CPH)对急性酒精中毒小鼠神经行为学及乙醇代谢酶活性的影响。方法:昆明种小鼠随机分为正常对照组,模型组,CPH低、中、高剂量组(1,2,4 g·kg^-1),醒酒一号葛根片组(XJGG 4 g·kg^-1),每组10只。给药组灌胃给予相应剂量的药物,正常对照组及模型组给予等量的0.5%羧甲基纤维素钠(CMC-Na),0.5h后,除正常对照组外,其余各组小鼠均灌胃给予56%乙醇(灌胃体积为5 g·kg^-1),在0.5,1,2h时分别检测各组小鼠神经行为学指标;同时使用酶联免疫吸附试验(ELISA)法检测各组小鼠肝组织中乙醇脱氢酶(ADH)、乙醛脱氢酶(ALDH)、微粒体乙醇氧化酶(EO)活性。结果:与模型组比较,CPH2,4 g·kg^-1剂量组在1,2h自主活动次数减少,在0.5~2h到达参照物的潜伏时间缩短,CPH4g·kg“剂量组在0.5~2h于转棒上的停留时间显著增加;此外,CPH2,4g·kg。能增加饮酒小鼠肝组织中ADH,ALDH,EO活性,其中以CPH4 g·kg^-1剂量组尤为显著。结论:CPH能增加乙醇代谢酶活性,抑制饮酒小鼠中枢神经系统兴奋性,改善乙醇所致的小鼠空间辨别障碍及共济失调现象,具有明显的解酒及中枢保护作用。Objective: To determine the detoxication effects of compound preparation of Hovenia dulcis Thunb (CPH) on the behaviors and activity of ethanol metabolizing enzymes in acute ethanol intoxication in mice. Methods : KM mice were randomly divided into groups, and treated with CPH ( 1,2, 4 g·kg^-1), XJGGK (4 g·kg^-1) or equal volume of 0.5% CMC-Na. At 0.5 h after the pretreatment, mice were intragastrically infused with ethanol (56% at 5 g·kg^-1). Then locomotor activity and the behaviors in Morris water maze and balance beam tests were assessed at 0.5, 1 and 2 h after ethanol ingestion. Meanwhile, the activities of ADH, ALDH and EO in liver tissue were tested by ELISA. Result: Compared with ethanol alone group, CPH 2 and 4 g·kg^-1decreased the locomotor activities at 1 and 2 h, and shorted the latency from 0.5 to 2 h. CPH 4 g·kg^-1 significantly increased the sustaining time in the rotarod from 0.5 to 2 h. Additionally, CPH 2 and 4 g·kg^-1 increased the activities of ADH, ALDH and EO in liver tissue in acute ethanol-intoxicated mice, especially in CPH 4 g·kg^-1 group. Conclusion: CPH can in- crease the activity of ethanol metabolizing enzymes, suppress irritability of the central nervous system, and improve spatial discrimination and ataxia phenomenon of acute ethanol-intoxicated mice. Thus CPH can alleviate alcoholism and has a protective effect on the central nervous system.
