血府逐瘀汤对动脉粥样硬化大鼠MMP-9及TIMP-1的影响

摘　　要：目的研究血府逐瘀汤对动脉粥样硬化(atherosclerosis,AS)大鼠动脉斑块基质金属蛋白酶9(MMP-9)及组织抑制物1(TIMP-1)的影响。方法将30只SD大鼠随机分为对照组、模型组、血府逐瘀汤组,对照组给予正常饲料,模型组给予高脂饲料,血府逐瘀汤组给予高脂饮食同时给予该药干预。12周后,处死大鼠,取主动脉免疫组化法观察主动脉斑块内MMP-9和TIMP-1的表达水平。结果血府逐瘀汤组MMP-9的表达较模型组明显减少(P<0.05),TIMP-1表达明显升高(P<0.05)。结论血府逐瘀汤可以通过下调动脉粥样硬化大鼠动脉斑块内MMP-9的表达,升高TIMP-1表达,起到防治动脉粥样硬化和稳定斑块的作用。Objective To investgate the effects of Xuefu Zhuyu Decoction on the expression of matrix metalloproteianse-9 and tissue metalloproteinase inhibitor-1 in aorta of atheroselerotic rats. Methods 30 SD rats were randomly divided into the control group model group and Xuefu Zhuyu Decoction group（each n --15）, the rats of the control group were given normal diet, that of the model group were given a high fat diet, the rats of Xuefu Zhuyu Decoction group were given a high fat diet and given treatment drugs. After 12 weeks, all rats were killed and expression of MMP-9 and TIMP-1 in atheroselerotic plaque were detected by using immunohistochemistry assay. Results The MMP-9 expression decreased significantly（P〈0.05）, and TIMP-1 expression increased significantly（P〈0.05） in the Xuefu Zhuyu Decoction compared with that of the model group. Conclusion The Xuefu Zhuyu Decoction can decrease the expression of MMP-9 and increase the expression of TIMP-1, it can attenuate atherosclerotic development in rats.
