摘 要:目的研究微小RNA Let-7与Lin28在散发性听神经瘤中的表达及二者之间的关系,探讨其对肿瘤进展可能的影响。方法收集18例经过术后病理证实的散发性听神经瘤组织作为实验组,7例正常胃迷走神经为对照组。采用RT-PCR、免疫组织化学染色方法,检测Let-7d和Lin28A/B mRNA的表达,以及Lin28蛋白的表达。结果 RT-PCR检测显示,Let-7dmRNA表达量在听神经瘤组织显著高于对照神经组织(P<0.001),Lin28A/B mRNA表达量在听神经瘤组织显著低于对照组织(PLin 28A=0.004,PLin28B=0.001)。在听神经瘤组织中,Let-7d与Lin28A/B mRNA的表达存在负相关关系(r=-0.671,P=0.002;r=-0.662,P=0.003)。免疫组织化学检测显示在听神经瘤与正常神经组织中均未发现Lin28蛋白阳性染色。结论在听神经瘤的生长及恶性转化抑制机制中,以微小RNA为主导的Let-7d/Lin28双向负反馈调节可能起重要作用。Objective To investigate the expression of miRNA Let- 7 and Lin28 and their association in sporadic acoustic neuroma in order to explore their role in the development of sporadic acoustic neuroma. Methods Real-time polymerase chain reaction (RT-PCR) was used to detect the mRNA expression of Let-7d and Lin28A/B in 18 sporadic acoustic neuroma tissues (experiment group) and 7 normal gastric vagus nerve tissues (control group). The relationship between the mRNA expression of Let-7d and Lin28A/B in sporadic acoustic neuroma tissues was analyzed. Immunohistochemical staining was used to detect the expression of Lin28 protein in the two groups. Results RT-PCR showed the mRNA expression of Let-7d was significantly higher in acoustic neuroma tissues than in control group (P〈0. 001), whereas the mRNA expression of Lin28A/B was significantly lower in acoustic neuroma tissues than in control group (PLin28A=0.004,PLin28B=0.001). The mRNA expression of Let- 7 d was negatively correlated with that of Lin28A (r= -0. 671,P=0. 002) and Lin28B (r= -0. 662,P=0. 003) in acoustic neuroma tissues. Immunohistochemical staining showed absent staining of I.in28 protein in both the sporadic acoustic neuroma and normal nerve tissues. Conclusion The double negative feedback regulation of miRNA Let-7d and I.in28 may play an important role in growth and malignant transformation inhibition of sporadic acoustic neuroma.
