摘  要:目的观察黑木耳多糖对大鼠脑胶质瘤生长的抑制作用,初步探讨其作用机制。方法提取并纯化得到黑木耳多糖用于后续实验。将27只SD大鼠随机分为3组:对照组、低剂量多糖组(100mg/kg)、高剂量多糖组(500mg/kg),观察各组肿瘤生长情况,于接种后30d处死各组大鼠,收集肿瘤标本测定肿瘤大小,荧光定量PCR及免疫组化检测肿瘤组织增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)的表达。结果低剂量多糖组、高剂量多糖组的肿瘤体积分别为(69.6±6.4)mm3和(62.2±4.9)mm3,均显著小于对照组(105.3±9.8)mm3(均P<0.01),且高剂量多糖组小于低剂量多糖组,差异具有统计学意义(P<0.01)。低剂量多糖组、高剂量多糖组肿瘤抑制率分别为33.9%和40.7%。PCNA荧光定量PCR和免疫组化结果显示低剂量多糖组、高剂量多糖组肿瘤组织PCNA表达减少,与对照组相比,差异均有统计学意义(均P<0.01),且高剂量多糖组的PCNA的表达量显著低于低剂量多糖组(P<0.01)。结论黑木耳多糖可显著抑制大鼠颅内胶质瘤的生长和抑制PCNA的表达,且高剂量多糖的抑制效果更为显著。Objective To examine the inhibitory effect of the black fungus polysaccharide on the growth of glioma in rats, and preliminarily explore its mechanism. Methods Black fungus polysaccharides were extracted and purified. C6 glioma cells were inoculated into the caudate nucleus of SD rate(n=27) which were randomly divided into three groups:control group,low-dose polysaccharide group (100 mg/kg), high dose polysaccharide group (500 mg/kg). The tumor growth was observed. Thirty days after inoculation,all the rats were sacrificed. Tumor xenografts were collected for measuring the volume of xenografts. The expression of proliferating cell nuclear antigen (PCNA) was detected by quantitative fluorescence PCR and immunohistochemis try. Results The tumor volume was (69.6--6.4) mm~ in low-dose group and (62.2± 4.9) mm3 in high dose group, signifi cantly lower than that in the control group which was (105.3+9.8) mma (P*~0.01 for all). The tumor inhibition rate was 33.9% and 40.7% in low-dose and high-dose groups, respectively. The immunohistochemistry and quantitative fluorescence PCR showed that the expression of PCNA was significantly decreased in low-dose and high-dose groups when compared with the control group with significant difference found (P〈0.01). Conclusion Black fungus polysaecharides can significantly in hibit the intracranial glioma growth in rats and the expression of PCNA,and the inhibitory effect of high-dose polysaccharides is more significant.
