摘  要:【目的】探讨转化生长因子(TGF-β1)诱导HL-60细胞凋亡后Smads蛋白的变化。【方法】用不同浓度的TGF-131处理HL-60细胞,采用MTT法检测细胞生长抑制率,流式细胞仪检测细胞凋亡,RT-PCR方法检测Smads蛋白。【结果ITGF-β1对HL-60细胞有增殖抑制作用,其抑制效应呈时间及剂量依赖性,10.4ng/mL TGF-β1增殖抑制作用较为明显;在细胞凋亡过程中,Smad3、Smad7表达呈逐渐升高趋势(P〈0.05),而Smad2、Smad4则无明显变化。【结论】TGF-β1可抑制HL-60细胞增殖并诱导其凋亡,且呈时间、剂量依赖性;Smad3、Smad7可能参与TGF-β1诱导HL-60细胞凋亡过程,其变化与HL-60细胞凋亡密切相关。[Objective] To explore the changes of Smads protein after HL-60 cell apoptosis induced by TGF-β1. [Methods] Different concentrations of TGF-β1 were used to treat HL-60 cells, the cell growth inhibition rate was detected by MTT method. Cell apoptosis was detected with flow cytometry and Smads protein was detected with RT-PCR. [Results] TGF-β1 had inhibitory effect on the proliferation of HL-60 cells, and the inhibitory effect was time- and dose-dependent. The effect of 10.4ng/ml TGF-β1 was more obvious. In the course of cells apoptosis, the expression of Smad3 and Smad7 increased gradually( P 〈0.05), but Smad2 and Smad4 had no apparent change. [Conclusion] TGF-β1 can inhibit HL-60 cell proliferation, and the inhibition effect is time- and d0se-dependent. Smad3 and Smad7 may be involved in HL-60 cell apoptosis induced by TGF-β1. Their changes are closely related with HL-60 cell apoptosis.
