摘 要:目的观察阿尔茨海默病(AD)模型小鼠视网膜各层细胞形态学改变。方法 6月龄AD模型小鼠及6月龄c57小鼠进行Morris水迷宫实验,检测小鼠的学习记忆能力。取小鼠眼球,制备成半薄切片,透射电镜下观察视网膜各层细胞形态学改变。结果 6月龄AD模型小鼠游行持续时间显著长于6月龄c57小鼠,差异有统计学意义(P<0.05);AD模型小鼠游行的路径长度和穿越次数均显著大于c57小鼠,但差异无统计学意义(P>0.05)。AD模型小鼠的视网膜光感受器细胞、外核层细胞、内网状层、内核层多层结构存在退行性改变。6月龄的AD模型小鼠脑内并未出现老年斑,但已经出现了学习记忆能力的下降。结论 AD模型小鼠视网膜多层结构存在神经退行性改变,这种改变早于颅内老年斑的形成。Objective To observe structural changes in every layer of the retina of Alzheimer disease (AD)model mice. Methods Six-month old Alzheimer disease model mice and c57 mice were executed Morris water maze test, to evaluate their learning and memory abilities. Eyes of mice, prepared into semi-thin sections were observed under transmission electron microscope. Results Six-month old AD model mice swam obviously longer before finding platform in water than those even-aged c57 mice, the difference was statistically significant ( P 〈 0.05 ). Six-month old AD model mice swam longer distance and experienced more crossing times than those of c57 mice with same age,but the difference was not statistically significant ( P 〉 0. ()5). Six-month old AD model mice had a decline in their ability of learning and memory. We found neuro-degenerative changes existed in multi-layer structures in retina, including retinal photoreceptors, the outer nuclear layer, inner plexiform layer, and inner nuclear layer. AD senile plaques in brain did not appear in these 6-month-old model mice, however, all the AD model rice had a decline in their ability of learning and memory. Conclusion The retinal nerve degeneration exists in AD model mice, which are even earlier than senile plaques formation in brain.
