猪繁殖与呼吸综合征病毒感染抑制肺脏抗菌肽PG-1基因表达

摘　　要：猪抗菌肽(Protegrin-1,PG-1)对革兰阳性菌、革兰阴性菌和产生耐药性的细菌具有抗菌作用,还可降低慢病毒亚科的传染性和抗囊膜病病毒感染。目的为了研究PG-1基因在猪体内的表达是否受到病毒感染的影响,我们设计了猪繁殖与呼吸综合征病毒攻毒试验,采用荧光定量PCR技术方法测定PG-1基因在肺脏和肠道的表达水平,分析猪繁殖与呼吸综合征病毒感染对PG-1基因表达的影响。结果表明,PG-1基因在肺脏和小肠组织中均有表达,与对照组比较,攻毒组肺脏和肠道中的PG-1基因的表达量均有所降低,肺脏中PG-1基因的表达水平下降显著(P<0.05),而小肠中PG-1基因下降不显著。结论分析认为,猪繁殖与呼吸综合征病毒感染抑制了抗菌肽PG-1基因在肺脏中的表达。As one of the antimicrobial peptides, Protegrin-1 has a broad-anti G^＋ and G bacteria spec- turn, and biological functions of anti virus infection. （Objective） To study the effect of virus on the ex-pression level of PG-1, （Methods） we designed a challenge trial using porcine reproductive and respiratory syndrome virus （PRRSV）. Lung and intestinal tissue were sampled for total RNA extraction and the ex-pressions of the PG-1 gene were measured by using absolute quantification PCR. The effect of PRRSV on the expression of PG-1 in lung and intestine was analyzed. （Result） The results showed that the PG-1 was expressed in lung and intestine, and the expressions of the PG-lwere decreased in both tissues. The ex-pression was decreased significantly （P〈0. 05） compared to the control. （Conclusion） It indicated that the infection of PRRSV suppressed the PG-1 expression in lung.
