摘  要:目的检测全身多发转移与无转移非小细胞肺癌患者血浆,筛选出与转移相关的血浆蛋白标志物。方法采用二维凝胶电泳(2-DE)技术分离多发转移及无转移24例非小细胞患者血浆蛋白质,PDQuest 7.1.0图像分析软件识别差异蛋白点,基质辅助激光解析电离飞行时间质谱及液相色谱-电喷雾串联质谱技术检鉴定差异蛋白。结果图像分析软件识别出12个差异蛋白点,基质辅助激光解析电离飞行时间质谱质谱鉴定10种差异蛋白,液相色谱-电喷雾串联质谱鉴定出其余两种差异蛋白。结论间α-胰蛋白酶抑制剂的重链4、载脂蛋白E在多发转移患者组血浆中表达上调,载脂蛋白A1、甲状腺素蛋白、AMBP蛋白、皮质类固醇结合球蛋白表达下调,提示这几种蛋白可作为非小细胞肺癌患者多发转移的血浆标志蛋白进行进一步研究。[ Objective ] To detect the plasma of two group patients with non-small-cell- lung cancer, find the metastasis-associated plasma protein markers. [Methods ] Use two-dimensional gel electrophoresis technical to sep- arate the protein which come from the plasma of two different group patients with non-small-cell -lung cancer: Systemic multiple transfer to no transfer. Screen differentially expressed proteins point with the PDQuest7.1.0 image analysis software. Use MALDI-TOF-MS and LC-ESI-MS/MS to identify the differentially expressed proteins spots. [Results] We screened 12 protein spots with the image analysis software and 10 protein spots was identified with the MALDI-TOF-MS, the remaining 2 spots was identified with the LC-ESI-MS/MS. [ Conclusions ] The expressed level of Inter-alpha-trypsin inhibitor heavy chain H4 and Apolipo-protein E were up regulated in muhiple metasta- sis patient group. Isoform 2 of Serum albumin. Transthyretin, Iimmunoglobulin lambda-chain Corticosteroid-binding globulin was down regulated in non transfer patients group. These proteins can be recognized as the protein markers which associated with muhiple metastasis in the plasma of non-small-cell-lung cancer patients.
