摘 要:目的:探讨乏氧环境对人牙髓细胞(hDPCs)增殖能力及碱性磷酸酶(alkaline phosphatase activity,ALP)活性的影响。方法:取第4代hDPCs分别在氧浓度为200mL/L(对照组)和10mL/L(乏氧组)的环境下进行培养;分别于培养后24h和48h,采用MTT法和PNPP法检测各组各时间点牙髓细胞的增殖能力及其ALP。结果:无论是乏氧组还是对照组,培养48h时的细胞增殖能力均明显高于其24h的增殖能力(P<0.05);而且同一时间点内乏氧组的增殖能力高于对照组(P<0.05)。两组中培养48h的ALP活性均明显低于其培养24h(P<0.05);但同一时间点内乏氧组的ALP活性明显高于对照组(P<0.05)。结论:乏氧环境能增强人牙髓细胞的增殖能力及碱性磷酸酶活性。AIM: To investigate the effects of hypoxia on the proliferation and alkaline phosphatase activity of human dental pulp cells ( hDPCs ). METHODS : hDPCs of passage 4 were incubated in normoxie condition ( 200 mL/L 02 ) and hypoxic condition ( 10 mL/L 02 ) for 24 h and 48 h trspevtively. Cell proliferation was determined by MTT assay. Alkaline phosphatase (ALP) activity was measured by PNPP method. RESULTS: Cell proliferation rates and ALP activity were significantly enhanced in hypoxia group as compared with those of normoxic group ( P 〈 0.05 ). CONCLUSION : Hypoxia can promote the proliferation and ALP activity of hDPCs.
